Tumornecrosisfactoralpha(TNF-alpha),alsoknownascachectinandTNFSF2,istheprototypicligandoftheTNFsuperfamily.Itisapleiotropicmoleculethatplaysacentralroleininflammation,immunesystemdevelopment,apoptosis,andlipidmetabolism(1,2).HumanTNF-alphaconsisitsofa35aminoacid(aa)cytoplasmicdomain,a21aatransmembranesegment,anda177aaextracellulardomain(ECD)(3).WithintheECD,humanTNF-alphashares97%aasequenceidentitywithrhesusand71%-92%withbovine,canine,cottonrat,equine,feline,mouse,porcine,andratTNF-alpha.TNF-alphaisproducedbyawidevarietyofimmune,epithelial,endothelial,andtumorcells(1,2).TNF-alphaisassembledintracellularlytoformanoncovalentlylinkedhomotrimerwhichisexpressedonthecellsurface(4).CellsurfaceTNF-alphacaninducethelysisofneighboringtumorcellsandvirusinfectedcells,anditcangenerateitsowndownstreamcellsignalingfollowingligationbysolubleTNFRI(2,5).SheddingofmembraneboundTNF-alphabyTACE/ADAM17releasesthebioactivecytokine,a55kDasolubletrimeroftheTNF-alphaextracellulardomain(6-8).TNF-alphABIndstheubiquitous55-60kDaTNFRI(9,10)andthehematopoieticcell-restricted80kDaTNFRII(11,12),bothofwhicharealsoexpressedashomotrimers(1,2,13).BothtypeIandtypeIIreceptorsbindTNF-alphawithcomparableaffinity(14),althoughonlyTNFRIcontainsacytoplasmicdeathdomainwhichtriggerstheactivationofapoptosis.SolubleformsofbothtypesofreceptorsarereleasedandcanneutralizetheBIOLOGicalactivityofTNF-alpha(15).

Chemotaxis elicited by Recombinant Human IL-8/CXCL8 (20&amp;nbsp;ng/mL) is neutralized (green line) by increasing concentrations of Mouse Anti-Human IL-8/CXCL8 Monoclonal Antibody (Catalog&amp;nbsp;# MAB208). Western Blot: iNOS Antibody [NB300-605] - Analysis of iNOS was performed by loading 20ug of RAW264 whole cell lysate untreated (left lane) or stimulated with LPS at 1ug/mL for 16 hours (right lane) and 10ul of PageRuler Plus Prestained Protein Ladder onto a 4-20% Tris-Glycine polyacrylamide gel. Proteins were transferred to a nitrocellulose membrane and blocked with 5% Milk in TBST for at least 1 hour. The membrane was probed with an iNOS Rabbit polyclonal antibody at a dilution of 1:1000 overnight at 4C on a rocking platform, washed in TBST, and probed with a Goat anti-Rabbit IgG (H+L) Secondary Antibody, HRP conjugate at a dilution of 1:1000 for 1 hour. Chemiluminescent detection was performed using SuperSignal West Pico.

Western Blot: NOD2 Antibody (2D9) [NB100-524] - Total protein from human THP1 and US-OS cells and mouse Raw264.7 cells was separated on a 7.5% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 2.0 ug/ml anti-NOD2 in 1% non-fat milk in TBST and detected with an anti-mouse HRP secondary antibody using chemiluminescence.
Immunocytochemistry/Immunofluorescence: NOD2 Antibody (2D9) [NB100-524] - A431 cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton X-100. The cells were incubated with anti-NOD2 (2D9) NB100-524 at a 1:200 dilution overnight at 4C and detected with an anti-mouse DyLight 488 (Green) at a 1:500 dilution. Actin was counterstained with Phalloidin 568 (Red) at a 1:200 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.
